HALO® Elevate C18 – Taking Separations to a Higher Level

Available now from Advanced Material Technologies is the new HPLC column phase HALO® Elevate C18.

Based upon proven Fused-Core® technology for excellent separation speed and highest chromatographic efficiency, the HALO® Elevate C18 provides the quality and reliability the HALO® brand is famous for. The HALO® Elevate C18 phase incorporates surface modified organo-silane technology for alkaline resistance, resulting in excellent stability under both high pH conditions and high temperature environments. With a wide operational use pH range of 2-12, HALO® Elevate C18 allows for robust method development and improved separations for basic compounds that may present problems at lower pH, such as poor peaks shapes, inadequate retention, or limited load tolerance. 

These new columns are designed and optimised to deliver excellent performance at high pH, as well as displaying the flexibility to work within the full range of operating conditions, allowing separation selectivity of acids, bases, neutrals and zwitterionic compounds.

Excellent Stability of HALO® Elevate C18 Over 500 Injections

Degradation of HALO® Elevate C18 silica particles is protected, which is demonstrated by thorough stability tests performed at elevated pH and temperatures.

The stability of a HALO® Elevate C18 column was tested using 10 mM ammonium bicarbonate at pH 10 and 60°C (> 20,000 column volumes, 42 hours of run time). The results below show that less than a 1% change in retention is achieved over 20,000 column volumes! HALO® Elevate C18 also demonstrated symmetrical peak shape, with no increase in back pressure during the 500 injection stability test.

The same 500 injection stability test was carried out under the same test conditions using competitor high pH columns, which were shown to fail in long term stability tests compared to the HALO® Elevate C18 column. The competitor columns displayed degradation, as well as peak splitting and peak fronting:

Performance You Can Rely On – The Speed of Fused-Core®

The performance of a HALO® Elevate C18 column was tested with the separation of Omeprazole, related compounds, and impurities. For best separation, omeprazole, with a pKa of 9.3, requires high pH. Performing this analysis at a pH of 10.7 at 60°C, a complete separation of 9 omeprazole related compounds and impurities was achieved in less than 4 minutes. The HALO® Elevate C18 was able to reduce run time by > 84% of the original USP method, saving significant time and solvent, while maintaining baseline resolution of all 9 compounds.

Excellent Lot-to-Lot Reproducibility

Lot-to-lot reproducibility is a key component in method development and validation. Six different lots of HALO® Elevate C18 were tested with a mix containing 5 basic compounds and 1 neutral compound, at pH 10.7. Results obtained showed the average %RSDs across all of the compounds was < 0.6%.

With a broad operational pH range of 2-12, HALO® Elevate C18 columns are perfect for method development, ensuring robust and reliable separations, especially for basic compounds. These columns are the ideal choice for high pH mobile phases, providing improved performance and superior results in challenging analytical applications.
